Dark Quencher-534 Phosphoramidites & Dark Quencher-579 Phosphoramidites are a non-fluorescent chromophore, and extensively used as the 3'-quencher moiety in a variety of FRET (Fluorescence Resonance Energy Transfer) in DNA detection probes. Such probes are primarily used in nucleic acid assays.
Advantages:
The advantages of using a Dark Quencher in a FRET probe are
- Low background fluorescence & higher dynamic range,
- Responsiveness to multiplex assays - because of a dark quencher having no secondary fluorescence.
- Comfort of synthesis of FRET probes with a Dark Quencher - because dark quenchers being resistant to degradation throughout the oligo deprotection step.
